Ropivacaine hydrochloride, CAS number 132112-35-7

Ropivacaine hydrochloride (RP-HCl) is a local anesthetic that is commonly used in medical practice. It is chemically classified as an amino amide local anesthetic and has the molecular formula C13H21NO3.HCl. It is used primarily for its analgesic properties during surgical procedures and for pain management.

In terms of its applications, RP-HCl has been studied for its effects on delayed cerebral vasospasm (DCVS) following subarachnoid hemorrhage (SAH) in animal models. Research has indicated that RP-HCl encapsulated in nanoliposomes (NLPs) can potentially improve neurological functions, reduce vasospasm, and mitigate vascular endothelial cell apoptosis. This suggests that RP-HCl NLP might have therapeutic value in managing the complications arising from SAH, although further clinical trials would be necessary to confirm these findings in human patients.

Additionally, RP-HCl has shown promise in reducing corneal sensitivity in horses when applied topically. It was found to be effective alongside lidocaine in decreasing corneal sensitivity without causing observable short-term adverse effects, making it a potential candidate for use in veterinary ophthalmology.

Precautions should be taken when administering RP-HCl, as with any medical treatment. These include ensuring correct dosing to avoid toxicity, monitoring for allergic reactions, and being aware of contraindications such as hypersensitivity to the drug or its components. In the context of systemic administration, care must be taken to prevent inadvertent intravascular injection which could lead to systemic toxicity.

Effect of ropivacaine hydrochloride and its dextroisomer (R)-ropivacaine hydrochloride on hERG currents using manual patch clamp method in HEK293 cells that stably overexpress hERG channel

Chinese Journal of Pharmacovigilance, 2024, Vol. 21, Issue (4): 391-396. DOI: 10.19803/j.1672-8629.20230326

To compare the effect of ropivacaine hydrochloride and (R)- ropivacaine hydrochloride on the human-ether-a-go-go-related gene(hERG) current in HEK293 cells that stably overexpress hERG potassium channels. Compared with dofetilide, ropivacaine hydrochloride has weak inhibitory effect on the hERG channel and (R)- ropivacaine hydrochloride has no obvious inhibitory effect on the hERG channel.

A Prospective, Single-Center, Single-Blind, Randomized, Confirmatory, Controlled Trial to Assess the Efficacy and Safety of PF-72 Combined with 0.75% Ropivacaine Hydrochloride in Patients Undergoing Single-Level Lumbar Discectomy

Int J Pain 2024; 15(1): 12-18 DOI: 10.56718/ijp.24-003

We conducted this study to assess the efficacy and safety of PF-72 (TGel Bio Co. Ltd., Seoul, Korea) combined with 0.75% ropivacaine hydrochloride in relieving the postoperative pain in patients undergoing single-level lumbar discectomy. PF-72 combined with 0.75% ropivacaine hydrochloride is an effective, safe drug delivery system.
